My DNAPrint Testing Saga



March 2003 and April 2003 Version 2.0 Tests - 71 Markers

I was physically sampled and tested twice with the DNAPrint 2.0 test in the spring of 2003. Once with a sample submitted via FamilyTreeDNA and the second time via a sample submitted directly to AncestrybyDNA. The results for the two tests in the spring of 2003 were exactly the same, i.e., 79% European and 21% Asian.

April 2004 Version 2.0 "Re-scored" Test

I requested a re-scoring from DNAPrint of my spring 2003 results because of their changing the computer algorithm in late 2003 to fix the African minority admixture sensitivity bug. Also I noticed coincident with that change more minority Native American results started being reported on the Genealogy-DNA-L List. I wanted to see if the algorithm change would affect my 2.0 test (71 marker) results. I received a version 2.0 "re-scored" CD in April 2004 which was supposedly a rescoring of my 71 marker genotype table of April 2003. But while the CD was marked with a 2.0 label it had my 173 marker genotype table on it from my January 2004 version 2.5 test. Something did not look right and I questioned AncestrybyDNA about it. Tony Frudakis of DNAPrint agreed to do a re-do of the "re-scoring". The CD I received in April 2004 later was found to be in error. But I will report here what was on it for continuity purposes. The April 2004 version 2.0 CD was supposedly a re-scoring of my 71 marker April 2003 test using the new iteration of 2.0 "scoring" algorithm introduced by DNAPrint in late 2003 to fix the minority African sensitivity bug in the algorithm. My minority admixture content which was 21% Asian in the two April 2003 tests was reported in the initial April 2004 CD as 12% Native American and 8% Asian. A very big change. As a result of my inquiry to DNAPrint/AncestrybyDNA about this puzzling CD with the wrong genotype table on it and dramatically different results, in June 2004 I learned that a human/clerical error had been made in the April 2004 "re-scoring" of my April 2003 version 2.0 genotype data. Someone used some subset of markers from my 173 marker genotype table from my January 2004 2.5 test instead of my actual 71 marker genotype table from April 2003. And since not all the markers in the version 2.0 genotype table are included in the version 2.5 genotype table the missing markers were marked as FL (failed) in the re-scoring effort by the programmer. The AncestrybyDNA.com programmer then ran the less than 71 marker sub-set of the 2.5 marker table with many markers FL'd through the new version 2.0 algorithm with the resulting confusing and erroneous 2.0 re-scored April 2004 CD being sent to me, i.e., a version 2.0 CD disk with a version 2.5 genotype table on it. I am now told by Tony Frudakis that the assayed results on that initial rescored April 2004 version 2.0 CD are definitely wrong. A new version 2.0 re-scored CD with the correct 71 marker genotype table of April of 2003 was received on 8 June 2004. The results thereon were: 79% European and 21% Asian. Thus the new version 2.0 computer scoring alogrithm did not change my results. Both the incorrect and correct re-scored version 2.0 (71 marker) test triangle plot and bar graph are shown below.

January 2004 Version 2.5 Test - 173 Markers

DNAPrint markets this test as a 175 marker test. But the genotype table in the Version 2.5 CD I received only lists 173 markers.

Note: DNAPrint Version 2.5 test is not simply more markers. DNAPrint has dropped some of the markers used in the March 2003 version 2.0 test. Only 48 markers of the 71 markers used in the March 2003 version 2.0 genotype set are still used in the January 2004 Version 2.5 173 marker genotype set. Also note that the version 2.5 test did not report any Native American minority content which was reported in the erroneous April 2004 version 2.0 "re-scored" CD discussed above in the version 2.0 "re-scoring" saga of the spring of 2004. The version 2.5 test only reported Asian minority content results which is consistent with the March and April 2003 version 2.0 tests and the corrected "re-scored" 2.0 CD received on 8 Jun 2004, but with tighter confidence bands and albeit with a much lower MLE.
